Mr. et Mrs. Smith sont des tueurs à gages qui travaillent pour deux organisations rivales. Ils se sont rencontrés dans des circonstances bien curieuses en Colombie, chacun travaillant pour une firme secrète sans que l'autre le sache, jusqu'au jour où ils se croisent sur une mission. La règle est simple : l'un des deux doit mourir, et ce sera l'autre qui le tuera…
